how much 100 line monofilament can I put on my 6/0 penn reel? 300 yards?

Probably 225-250 yards. But why straight 100lb on the whole spool? I assume this is for surf fishing right?

You'll be able to get the bait out further which is where the big shark live if you spool it up with 50lb mono or 65-80lb braid and then splice a 100 yard piece of 80lb mono to your main line. The 80lb is what will be in the sand and it's a lot more abrasion resistant than the 50lb mono or braid. I have all my 6/0 spooled with 50lb Ande and then a 100 yards of 80lb Ande. This is just my two cents.
